(E). Exogenous antioxidants can be produced synthetically. However, these synthetic compounds are carcinogenic. The compounds expected to have non-carcinogenic antioxidant activity are exopolysaccharides (EPS). One of the potential EPS produces as antioxidants is Lactobacillus plantarum bacteria which is a class of lactic acid bacteria (LAB). This study to characterize the monomer types of EPS produced by Lactobacillus plantarum by high performance liquid chromatography (HPLC) method with a standard comparative of glucose and mannose and antioxidant activity of EPS in vitro against sheep blood cells experiencing oxidative stress induced by t- BHP. The antioxidant activity of EPS was determined by three test parameters, the measurement of malondialdehyde (MDA) level using thiobarbituric acid reactive substances (TBARS) test method, the measurement of catalase activity was performed by using peroxide solution in phosphate buffer solvent, and measurement of superoxide dismutase (SOD) adenochrome assay. The result of characterization of EPS with HPLC showed that the monomers were glucose sampling by observation of sample retention time of 5.205 min close to BP glucose retention time (5,339 min) but not close to retention time of mannose (5,691 min). Measurement of antioxidant activity based on EPS concentration variations of 10, 20, and 40 μg / mL. Measurement results of MDA activity between 8.0-10.5 nmol / mL. The result of SOD activity measurement is between 12,0-20,0 unit / mL. Results of measurement of catalase activity between 350.0-500.0 units / mL. Statistical tests show that EPS administration of sheep blood cells with oxidative stress can decrease MDA levels and increase SOD activity and catalase. This proves EPS with a glucose-forming monomer having antioxidant activity.
